When submitting a manuscript, is it all right to use Courier New 10 point, or do you have to use Courier New 12 point?

I'd do 12pt. Make it as easily readable as possible.

Unless the editor/publisher has specifically asked for work to be in a specific fontface or of a certain size, I think you're fine. BUT...as a personal observation, 10pt is pretty frickin' small for long reads. Unless there's an overriding reason for using 10 (like you're a starving artist and you can't afford the extra paper) I'd use 12.

Almost all guidelines request 12-point font, Courier or Times New Roman.

You wouldn't want to make a poor agent or editor's eyes bleed, would you? :D
