I've been going to comic-con for years. (My husband used to shoot their commercials back in the day.) It's gotten incredibly huge and overwhelming. Last year I think there were 60k people attending and lines around the block. My husband is going, but I'm still on the fence. Here's some advice:
Bring water and pocket snacks. The convention center concessions are ridiculously expensive.
Wear comfortable shoes (and maybe some elbow pads). The con takes up the entire convention center. The dealers room is the WHOLE bottom floor.
Give yourself lots of time to get from place to place.
Be sure to leave the convention center and go into the Gaslamp district for at least one meal. There are a ton of wonderful restaurants crammed into just a few city blocks.
Parking is horrible during the con. Take public transportation when possible. The trolley has a stop just across the street from the convention center. (I park for free at old town and take the trolley in.)
There is a sort of mezzanine with a big plate glass window that looks down on the dealers room (North-west side I think?). There are some concession stands and tables there. It is the absolute best place to take a break from the throng and people-watch.

Great advise. Last year there was about 12000 folks in the hall over 5 days. This year should be more.
I have gone about 3 times before and this is what I found.
- Go to Ralphs (walking distance to the hall) and get a nice sandwich or make one at home DO NOT BUY CONVENTION FOOD
- if your a pro, get a pro badge. You don't have to wait in the super line. I know folks that waited in line for an hour, after the hall opened to get it. Pro's get in 15 min earlier and of course being an exhibitor rocks.
- The Atrium where the free-be table (up stairs) is a better place to meet.
- on busy days, don't try to cross the hall. Exit into the lobby and walk down and re-enter. Trust me, the crowd gets nasty on Fri-Saturday and just trying to make it past the Star Wars fans is enough to make you faint.
- When it's busy it is a good time to retreat to the back areas of the hall, away from the buzz booths. Small press is where some of the great new stuff is, and check out artist alley too.
