Does this work better with or without quotes?

I don't really like to see quotes used for emphasis. I prefer italics (or, in a pinch, boldface). In this case, I'd use italics.

So.....a brawny female...

What makes you so sure she was a she? I have seen some males without shirts that could still pass for female. I just wondered what made you think this was a female? Was she really endowed? (Just me and my wondering mind.)

I would agree with the italics, instead of quotes. And I have to ask -- how did my stepsister get into your novel?
