Yes, proper manuscript format for all submissions. It's easier to read, and if you're very lucky (or good) it leaves space for notes.

Standard manuscript format:
1 inch margins all around (top, bottom, left and right)
12 point font (times new roman or courier - I like TNR, but either is fine)
Double spaced
Either a cover page or on the first page:
your name
contact info
word count
Title
genre
Then, for all the rest of the pages, add a header to all the rest of the pages that is: Your Last name/Title/page number in the upper right hand corner of the page.
Start each new chapter about a 1/4 of the way down the page.

Actually I lied. This is a slightly different question, but about that first page: If the agent doesn't like attachments and wants everything in the body of your email, should you still include that first page? And if so, should you just basically put a space between that info and your first chapter?
I'd look at the website of the people you are sending it to. One of my publishers wants double spacing the other 1.5. If they have specified something other than double it would be unwise to give them something else. They might also ask for other specific formatting. But if there is no instruction given, definitely double spacing.
