When you get reply from a query saying 'send me the full/first fifty', do you normally just reply back to that original email with the manuscript attached? Or do you reply, but change the subject line to something like 'manuscript request for blahblahblah'. Or do you perhaps send out a completely new email?

Just curious. I have been doing a simple reply with an added attachment, and now am wondering if it might get lost in the query slush.

Reply to the original email so the agent can see all prior discussion where he/she requested. Change the subject line to just say "Requested Materials: TITLE" rather than a whole bunch of Fwd: RE: mess.

At the point of giving a partial/full, the agent is expecting to hear from you, and so you can attach a file to the email at that point (unless of course you are told not to do that).
