Is there a way I can search barnesandnoble.com to see if a local store has a book in stock?

The last time I checked, B&N didn't offer that option. Borders does.
You can call your local B&N and ask about the book though.

Thanks! I'll check out the Borders site. Borders is closer to my house, anyway.

They had that function available several months ago, but I tried to use it again recently and couldn't find it. It's either gone or well hidden.

As far as I recall when I did use it, I searched on the title first, then once I was looking at the information for the specific book, there was an option to check in-store availability. I put in my zip code and it showed which nearby stores had it in stock.

Yep. When you click on a book, there's a a "FIND & RESERVE AN IN-STORE COPY" box (on the right, for me). Entering your ZIP code will bring up a list of nearby stores showing In Stock or Out of Stock as applicable.
