Writing to an agent's assistant

I've found an agent I'd like to contact but the only email I can find is for her assistant. What's the best way of wording my initial query? I don't want to come across as I don't want to talk to you. You're just the servant. Get me the important person.

I say! I say! You there, manservant. Inform your master I desire to communicate with him. There, run along, there's a good chap.

I'd check first to make sure that the query guidelines say use that email address when querying. The agent should have how they want you to contact them posted somewhere, either on their website or blog or in a Writers' Market type book.

If that's the address they say to use, address it to the agent. The assistant is there to "open the mail" so to speak.

I sent it yesterday and got a reply from the assistant this morning. They're now the assistant to another agent who doesn't deal with fiction.

Bugger.
