I sent off my first two submissions to Highlights for Children, an activity rhyme and a story. Received a "scales" card for the activity rhyme (still waiting to hear on it) and a rejection on the story, *but* I got a hand written note on my story manuscript explaining the rejection and asking to see more of my work.
Does that qualify me to put "requested manuscript" on the envelope of my next story submission to Highlights? Or is that pushing it? :D

Oh, that would definitely be pushing it. An editor won't appreciate opening an envelope saying "Requested Submission" only to find out that she didn't request it at all.
The editor's already given you a green signal. Run with that. Send her more stuff. But I wouldn't advise marking it as Requested.

Thanks, I kind of figured that. :)
So, in my cover letter, I thank her for the kind comments and say I'm sending along a new story per her suggestion? Is that the best way to handle it?

That would be good. The Highlights folks are very nice and if you give a thank you in your cover letter, you should -- at least -- get another note back instead of the horrors of check-list rejection.
